Hi!

If a player describe a humanity-loss activity in the kicker (case in point: "I just sacrificed my best friend killing him on an altar to the dark gods") he has to roll for humanity loss or it's considered something that happened before playing and there is no roll?

You do make Humanity checks for the Binding, pre-play. There are no other rolls for pre-play actions.
